ABSTRACT The preservation of banana genetic material is usually performed through seedlings. However, most banana cultivars do not produce seed and are propagated vegetatively. Therefore, cryopreservation is a feasible technique that allows the preservation of banana genotypes indefinitely. For the success of cryopreservation protocols, the selection of cryoprotectants and pre-freezing techniques are important factor. The aim of this study was to evaluate the efficiency of vitrification and droplet vitrification for the cryopreservation of Hancornia speciosa shoot tips. The shoot tips were subjected to four different periods of exposure (15, 30, 45 and 60 min.) to plant vitrification solution 2 (PVS2) before plunging into liquid nitrogen. We evaluated the regrowth of H. speciosa shoot tips that were cryopreserved by the classical vitrification technique and by the droplet vitrification technique. Abstract Psychotria ipecacuanha is a medicinal plant endangered in Colombia, due to the indiscriminate exploitation suffered several decades ago for its therapeutic importance. Its great medicinal value is due to emetine production and its derivatives, mainly in the roots. Given these concerns, the aim of this research was to evaluate different parameters for in vitro conservation of P. ipecacuanha by a limited plant growth, using as in vitro explants plant nodes. In vitro conservation of diploid banana accessions Scientia Agricola

A protocol for the in vitro conservation of diploid banana accessions based on lower temperature for culture environment was developed. Twenty four genotypes, four clones per genotype were studied. After disinfection, shoot tips (0.5 cm diameter x 0.5 cm height) were established in vitro and submitted to multiplication induced by benzylaminopurine. Twelve explants (0.6 cm diameter x 1.2 cm height) from each accession were transferred to the MS medium supplemented with 20 g L-1 sucrose and 2 g L-1 Phytagel, without growth regulators, under light intensity of 2000 lux, 16-hour photoperiod, at temperatures of: 17 ± 2ºC, 22 ± 2ºC and 26 ± 2ºC. Influence of substrates and in vitro preconditioning treatments on ex vitro acclimatization of Arachis retusa PAB

The objective of this work was to evaluate the influence of substrate and preconditioning treatments on the acclimatization of in vitro plants of Arachis retusa. Plants were transferred to Plantmax or sand, and fertilized with Hoagland's nutrient solution. Plants maintained in sand, with or without fertilizer, showed the highest survival rates. In order to evaluate the influence of in vitro preconditioning treatments, stem segments were cultured on MS medium supplemented with different sucrose concentrations. The highest survival and developmental rates were observed in plants from two accessions cultured on MS supplemented with 1.5% and 3% sucrose. Flowering and fruit production were observed after five months.

In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. At proliferation stage, the maximum number of new leaves (9.6) were produced on the medium containing 4 µM BAP + 0 µM GA3, whereas the maximum number of axillary shoots (4.1) were observed in the medium containing 4 µM BAP + 3 µM GA3. Short-term storage in vitro and large-scale propagation of grapevine genotypes PAB

The objective of this work was to evaluate the large-scale propagation of grapevine genotypes after short-term storage in vitro. Microshoots from ten grapevine genotypes were used. The following storage temperatures were evaluated: 10, 20, and 25°C. After short-term storage, the shoots were propagated in up to five successive subcultures, to assess the large-scale propagation of the germplasm maintained under conditions of minimal growth. The propagated shoots were rooted in different concentrations of indolbutiric acid (IBA) and acclimatized in greenhouse. Survival of sugarcane shoot tips after cryopreservation by droplet-vitrification PAB

The objective of this work was to evaluate the phytotoxicity of a plant vitrification solution (PVS2), and the survival of shoot tips of the sugarcane variety SP716949, after cryopreservation by droplet-vitrification. Shoot tips were precultured for 24 hours in MS medium containing 0.3 mol L-1 sucrose, and exposed to PVS2 for 0, 20 or 30 min. Shoot tips were then immersed in liquid nitrogen. Thawing was fast in concentrated sucrose solution (1.2 mol L-1). PVS2 is a nontoxic to shoot tips, which in turn are sensitive to liquid nitrogen. The best results occurred when shoot tips were maintained for up to 20 min in PVS2 solution, before freezing, with 20% survival.
